ABSTRACT

Objective:To study the association between metabolic syndrome (MS) and Chinese healthy eating index (CHEI), and to identify the beneficial or adverse effects of diets on MS in a community population of Shanghai. Methods:Based on the inclusion and exclusion criteria, the data of 4 856 subjects from a community in Shanghai was collected by interview. Total CHEI score and its component score were calculated based on the frequency of food consumption. Physical examination and blood biochemical tests were used to diagnose MS. The logistic regression model was used to analyze the relationship between MS or relative indexes and the total CHEI score or its component score. Results:The study showed the overall prevalence of MS was 24.71%. There were significant differences between MS group and controls (P<0.05) in age, BMI, waist circumference, fasting blood glucose, triglycerides, total cholesterol, systolic blood pressure, diastolic blood pressure, glycated hemoglobin, high-density lipoprotein and energy intake. After adjusting for potential confounders, it showed that the higher total CHEI score, the lower risk of central obesity, increased diastolic blood pressure, increased glycated hemoglobin, hyperglycemic and MS (P<0.05). The increase in component score of potatoes, milk, beans, dark vegetables, fruits, oil and sodium reduced risk of obesity, hypertension, hyperglycemia, dyslipidemia and MS (P<0.05). Conclusion:The quality of healthy diet (CHEI score more than 80) and increased intake of specific dietary components (potatoes, milk, beans, dark vegetables, fruits) reduce the risk of MS effectively.

ABSTRACT

Objective:To investigate the prevalence of metabolic syndromes (MS) and MS scores as well as related health behavior factors in Zhongshan Community, Songjiang, Shanghai. Methods:A total of 6 802 residents aged 20-74 years old in Zhongshan Community were selected. Face-to-face survey and body measurement were used to collect information such as MS-related behavioral factors (including smoking, alcohol intake, exercise and diet) and to determine the MS scores. MS scores were divided into 6 levels. Ordered logit model was used to analyze the factors related to MS score, and logit model was used to analyze the factors related to MS. Results:The prevalence of 6 metabolic syndrome scores in the sample population were 13.5%, 24.3%, 25.1%, 19.7%, 12.3%, 5.1%, respectively. The prevalence of MS was 37.0%. The related factors of high MS score in male were advanced age, alcohol intake and tea drinking, while the related factors of high MS score in female were advanced age and previous smoking. The related factors of MS was alcohol intake in male while female with advanced age had higher risk in developing MS. Conclusion:The prevalence of MS in Zhongshan Community is relatively high, which has become one of the important public health problems in this community. Attention should be paid to the elderly men who drink alcohol and tea, and aged women who have ever smoked.

ABSTRACT

AIM: To observe the therapeutic effect and safety of improved extra panretinal photocoagulation ( E - PRP) in the treatment of high risk proliferative diabetic retinopathy (hsPDR). METHODS: A total of 88 consecutive cases (102 eyes) with hsPDR were diagnosed by fundus fluorescein angiography(FFA) from February 2011 to December 2014 in our hospital . Fifty two eyes had been treated by improved E - PRP with 532nm frequency - doubled laser. Fifty eyes had been treated by standard PRP. All cases were checked by FFA and fundus photocoagulation every 3mo. Patients with persisting neovascularization or non perfusion area were treated with laser again. All cases were followed up 6-36mo. RESULTS: The postoperative visual acuity had no statistical difference between two groups ( P > 0. 05). In improved E - PRP group, retinal non perfusion area and neovascularization disappeared in 35 eyes ( 67%). Effective rate was 88%. Six eyes (12%) underwent pars plana vitrectomy because of vitreous hemorrhage, fiberosis and stretched retinal detachment. In standard PRP group, retinal nonperfusion area and neovascularization disappeared in 23 eyes ( 46%). Effective rate was 66%. Seventeen eyes(34%) underwent pars plana vitrectomy because of anterior retina bleeding or vitreous hemorrhage. The rate of neovascularization disappeared and effective rate had statistical difference between two groups (P CONCLUSION: It is a safe and effective methods to treat hsPDR by improved E-PRP and it was more effective than traditional PRP.

ABSTRACT

By a orthogonal experiment, the influence of different ratio of phosphorus and potassium fertilizers on imperatorin, isoimperatorin and psoralen contents and yield of Glehnia littoralis were studied. The results showed that root dry weight and the yield of G. littoralis increased when reasonably applied phosphorus fertilizer combined with potassium fertilizer within a certain range. And the influence of phosphorus fertilizer was greater than that of potassium fertilizer. The optimal value of root dry weight and yield achieved at both P2O5 360 kg x hm(-2), K2O 270 kg x hm(-2) and P2O5 360 kg x hm(-2), K2O 180 kg x hm(-2). The effects of different phosphorus and potassium treatments on the content of imperatorin, isoimperatorin and psoralen in G. littoralis were determined, which shows that the content increased with the moderate increase of phosphorus and potassium. And the effects of phosphorus fertilizer were more significantly. The isoimperatorin content achieved the largest value at P2O5 360 kg x hm(-2), K2O 270 kg x hm(-2), also a larger content of imperatorin and psoralen. The imperatorin content is the largest when applied P2O5 360 kg x hm(-2), K2O 180 kg x hm(-2), and the isoimperatorin content was higher as well. So that the treatment of P2O5 360 kg x hm(-2), K2O 180 kg x hm(-2) are suitable for promote to the agricultural production, which could improve the quality and yield of G. littoralis.

ABSTRACT

Objective To establish and optimize a sensitive and specific quantitative realtime polymerase chain reaction(PCR)method for detection of hepatitis B virus covalently closed circular DNA(HBV cccDNA)in liver tissue. Methods Specific primers and probes were designed to detect HBV DNA(tDNA)and cccDNA. A series of plasmids(3.44 × 100-3.44 × 109 copies/μl)containing a full double-stranded copies of HBV genome(genotype C)were used to establish the standard curve of real-time PCR. Liver samples of 33 patients with HBV related hepatocellular carcinoma(HCC), 13 Chronic hepatitis B patients(CHB)and 10 non-HBV patients were collected to verify the sensitivity and specificity of the assay. A fraction of extracted DNA was digested with a Plasmid-Safe ATP-dependent Dnase(PSAD)for HBV cccDNA detection and the remaining was used for tDNA and β-globin detection. The amount(copies/cell)of HBV cccDNA and tDNA were measured by a real-time PCR, using β-globin housekeeping gene as a quantitation standard. Results The standard curves of real-time PCR with a linear range of 3.44 × 100 to 3.44 × 109 copies/μl were established for detecting HBV cccDNA and tDNA, and both of the lowest detection limits of HBV cccDNA and tDNA were 3.44 × 100 copies/μl. The lowest quantitation levels of HBV cccDNA in liver tissues tested in 33 HBV related HCC patients and 13 CHB patients were 0.003 copies/cell and 0.031copies/cell, respectively. HBV cccDNA and tDNA in liver tissue of 10 non-HBV patient appeared to be negative. The true positive rate was increasing through the digestion of HBV DNA by PSAD, and the analytic specificity of cccDNA detection improved by 7.24 × 102 times. Liver tissues of 2 patients were retested 5 times in the PCR for detecting cccDNA and the coefficience of variations on cycle threshold (Ct)were between 0.224%-0.609%. Conclusion A highly sensitive and specific quantitative real time PCR method for the detection of HBV cccDNA in liver tissue was established and could be used for clinical and epidemiological studies.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the clinical value of 3D visualization method for simulating percutaneous transcatheter closure of atrial septal defect (ASD).</p><p><b>METHODS</b>3D volume render method was used for visualizing ASD and surrounding structures and 3D modeling method was applied for simulate the shape of occlusion device. The size and the distance between the lower edge of device and atria-ventricular valve of simulation occluder and actual selected atrial septal occluder (ASO) were compared in 30 patients underwent successful transcatheter closure of ASD.</p><p><b>RESULTS</b>The location, geometry, size, extent of ASDs in children were displayed in 3D visualization. No significant difference was found between simulation occluder and ASO size measured from left atrium [(26.07 +/- 5.32) cm vs. (25.91 +/- 5.32) cm] and right atrium [(22.13 +/- 5.31) cm vs. (22.08 +/- 5.26) cm, all P > 0.05]. The distances from simulation occluder to mitral valves [(5.76 +/- 2.39) cm] and to tricuspid valves [(8.25 +/- 2.40) cm] were similar as ASO to atria-ventricular valves [(5.61 +/- 2.26) cm and (8.02 +/- 2.48) cm, respectively, all P > 0.05].</p><p><b>CONCLUSIONS</b>The simulating percutaneous transcatheter closure of ASD by 3D visualization technique could be a helpful noninvasive approach for ASO selection before the procedure of transcatheter occlusion of ASD.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the value of 3-dimensional CT in the treatment of developmental dislocation of hip (DDH).</p><p><b>METHODS</b>From 2003.6 to 2007.6, the femoral neck anteversion (FNA) and morphology of acetebulum in 53 patients with DDH (61 hips) were studied by three-dimensional CT imaging. Among the patients, 12 patients were male and 41 patients were female, ranging in age from 3 to 16 years (mean 5.6 years). Thirty-four patients had dislocation in left hip joint, 11 patients had dislocation in right hip joints, and 8 patients had double dislocations. The patients were treated with Pemberton or Chiari acetabuloplasty and femoral osteotomy. After operation all the cases were obeserved by 3-dimensional CT again. The femoral neck anteversions were measured and the shapes of new acetabulum were observed.</p><p><b>RESULTS</b>Among 61 hips of DDH, the maximum femoral neck anteversion was 90 degrees. The minimum was 35 degrees and the average was (45.6 +/- 11.4) degrees. Among 45 normal hips, the average femoral neck anteversion was (23.5 +/- 10. 2) degrees. The acetabulum were dysplastic according with what were found in the operation. After operation the femoral neck anteversions decreased and averaged (15.6 +/- 5.8) degrees. The femoral head containment improved.</p><p><b>CONCLUSION</b>The advantages of 3D CT scan includes manifestation of acetebular morphology, correct measurement of femoral neck anteversion and evaluation of operative procedure and efficacy. The 3-dimensional CT method is deserved to use widely in the treatment of developmental dislocation of hip in children.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the status of occult hepatitis B virus infection in chronic viral hepatitis patients with non-A to E hepatitis virus infection and explore the diagnostic value of fluorescence quantitative polymerase chain reaction (FQ-PCR) technique for occult hepatitis B virus infection.</p><p><b>METHODS</b>The amount of HBV-DNA in serum and liver tissue from 57 patients with non-A to E hepatitis virus infection who were diagnosed as chronic viral hepatitis by Menghini method liver biopsy were detected by using FQ-PCR technique, then the relation between the viral load of HBV DNA in liver tissue and hepatic inflammatory activity were analyzed.</p><p><b>RESULTS</b>Thirteen (22.81%), 22 (38.60%) patients were positive for HBV DNA in serum and liver tissue, respectively. The positive rate and the level of HBV DNA quantity in liver tissue were significantly higher than those in serum; HBV DNA was found positive in both serum and liver tissue in 13 cases, negative in both serum and liver tissue in 35, positive in liver tissue but negative in serum in 9, and in none of the cases HBV DNA was positive in serum but negative in liver tissue (P < 0.01). The logarithmic value of HBV DNA from 13 patients in liver tissue and in serum was respectively: (6.62 +/- 1.21) copies/g vs.(4.03 +/- 1.06) copies/ml, P < 0.01. The hepatic lesions of all HBV DNA positive patients were active pathologic changes, but the level of HBV DNA in liver tissue was not significantly correlated with the grade of hepatic inflammation activity (P > 0.05).</p><p><b>CONCLUSION</b>Occult HBV infection is the etiology of part of the chronic viral hepatitis patients with non-A-E hepatitis virus infection. Missed diagnosis will occur if diagnosis of hepatitis B is only based on detection of serum HBV markers. It is useful for improvement of the diagnostic level of HBV infection via detection of HBV DNA quantitatively in serum especially in liver tissue of chronic viral hepatitis patients with non-A-E hepatitis virus infection by using FQ-PCR technique. The chronic viral hepatitis patients with occult HBV infection should be also given effective anti-viral therapy.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To examine the antigenicity of hepatitis C virus (HCV) F protein and investigate serum prevalence of anti-F in HCV-infected patients.</p><p><b>METHODS</b>Eleven pairs of overlapping primers were used to synthesize the full-length HCV f gene, from which the truncated HCV f65 gene fragment was amplified by PCR. HCV f65 gene was then cloned into pET32a(+), and transformed into E. coli strain Plyss (DE3). This recombinant E.coli was induced by IPTG for the production of HCV F65 protein. The expressed HCV F65 protein, purified by Ni-NTA agarose, was further used in ELISA to detect serum anti-F, and to immunize rabbits for making polyclonal anti-F. The rabbit polyclonal anti-F was purified by Staphylococcus aureus protein A agarose.</p><p><b>RESULTS</b>After recombinant pET32a(+)-f65 was constructed successfully, HCV F65 protein was expressed and purified. The purified HCV F65 protein was used as a capture antigen in ELISA to detect serum anti-F in HCV infected patients (n = 30). The result showed that the mean A450 value and the positive rate of serum anti-F were 0.125+/-0.061 and 63.3%, respectively. The rabbit-derived polyclonal anti-F reacted specifically with HCV F65 protein, of which the titer was 1:30,000.</p><p><b>CONCLUSION</b>Our expressed HCV F65 protein is of antigenicity, and can be used to determine serum anti-F. Anti-F IgG does exist in the sera of the HCV-infected patients. Moreover, the rabbit-derived polyclonal anti-F can be used to detect HCV F protein.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effect of hepatitis C virus (HCV) superinfection on the short-term and long-term hepatic pathological changes in patients with chronic hepatitis B (CHB).</p><p><b>METHODS</b>HCV-RNA of twice corresponding period serum samples was detected via reverse transcription polymerase chain reaction assay from 230 patients with CHB for whom liver biopsy was performed at an interval of 0.5-15 years, respectively. The hepatic pathological changes of the patients with CHB who were serum HCV-RNA positive at the beginning of observation and persistently positive between the starting and ending of observation were respectively compared with those of serum HCV-RNA negative and persistently negative patients.</p><p><b>RESULTS</b>41 patients (17.83%) were positive for serum HCV-RNA at the beginning of observation. There were significant differences in the severity of hepatic inflammatory activity grade and fibrosis stage between serum HCV-RNA positive and negative patients with CHB (P < 0.05). Twenty-nine patients were persistently positive for serum HCV-RNA in the beginning and end of observation. Compared with persistently negative patients who were 116 patients selected from the above-mentioned 230 patients and they were comparable with HCV-RNA persistently positive patients in mean follow-up time, age and sex, the long-term progression of hepatic inflammatory activity grade and fibrosis stage in persistently positive patients were more speedy (P < 0.01).</p><p><b>CONCLUSION</b>HCV superinfection worsens the hepatic pathological changes of patients with CHB and speeds up its progression.</p>

ABSTRACT

Strain QJ16,one of some methanotrophic bacteria isolated from soil samples utilizing the separate device designed by our laboratory;was studied in detail and identified as one of the Genus methylomonas through 16S rDNA sequencing and comparison. The culturing and reaction conditions for methane utilization of this strain were investigated. The results indicated that the optimal conditions for the growth of strain QJ16 were methane gas as carbon source and the coexistence of NH4Cl and KNO3 as nitrogen source,the cultivation temperature of 30℃ and medium initial pH 6~7,and the optimal Cu2+ concentration of 15 ?mol/L.
